1. ¿¬±¸ºÐ¾ß ¹× ³í¹®³»¿ë ¼Ò°³

¿ì¼± Á¦°¡ ÁÖ·Î »ç¿ëÇÏ´Â electron tomography¿¡ ´ëÇØ ¸»¾¸µå¸®°Ú½À´Ï´Ù. Tomography¿¡¼­´Â °üÂû´ë»óÀ» Åõ°úÇÏ´Â wave³ª particleµéÀ» ¿©·¯ °¢µµ¿¡¼­ ½î¾Æ¼­ Åõ¿µµÈ »óµéÀ» ¾òÀº ÈÄ, ±× »óµéÀ» ÄÄÇ»ÅÍ ¾È¿¡¼­ À籸¼ºÇÏ¿©, °üÂû´ë»óÀÇ ³»ºÎ±¸Á¶, Áï ´Ü¸éµµ¸¦ ¾ò¾î³À´Ï´Ù. MRI¿Í CT¿¡¼­´Â °¢°¢ radio frequency ¿Í X-ray (ȤÀº ion beam)°¡ »ç¿ëµË´Ï´Ù.

Electron tomography´Â electron beamÀ» ÀÌ¿ëÇÏ´Â tomographyÀÇ ÇÑ Á¾·ù·Î, ¹ßÀüµÈ ÇüÅÂÀÇ transmission electron microscopy (TEM)ÀÔ´Ï´Ù. °íÀüÀûÀÎ TEM¿¡¼­´Â sampleÀ» 60~80 nm µÎ²²ÀÇ sectionÀ¸·Î ÀÚ¸¥ ÈÄ ±× section¿¡ electron beamÀ» ½î¾Æ¼­ Æò¸é¿¡ Åõ»çµÈ »óÀ» ¾ò½À´Ï´Ù. TEM sectionÀº 3Â÷¿ø ¹°Ã¼ÀÌÁö¸¸ 2Â÷¿øÀÎ Æò¸é¿¡¼­ °üÂûÀ» Çϱ⠶§¹®¿¡ ¸¹Àº ±¸Á¶ Á¤º¸, ƯÈ÷ electron beam°ú ÆòÇàÇÑ ¹æÇâÀÇ À§Ä¡ Á¤º¸´Â ¸ðµÎ ¾ø¾îÁö°í, ¶Ç ÇØ»óµµµµ ¸¹ÀÌ ¶³¾îÁö°Ô µË´Ï´Ù. ¹Ý¸é electron tomography¸¦ »ç¿ëÇϸé 60 nmÀÇ sectionÀÌ 30°³ÀÇ ¼ø¼­´ë·Î Á¤¿­µÈ 2 nm µÎ²²ÀÇ ´Ü¸éµµµé (tomographic slices)·Î ³ªÅ¸³³´Ï´Ù. °¢°¢ÀÇ ´Ü¸éµµµé¿¡¼­ ÇØ»óµµ´Â 4 nm Á¤µµ·Î ±âÁ¸ÀÇ ÀüÀÚÇö¹Ì°æ»çÁøµé¿¡¼­´Â È®½ÇÇÏÁö ¾Ê´ø ±¸Á¶µéÀÌ Á¤È®È÷ °üÂûµË´Ï´Ù (¾Æ·¡ ±×¸² ÂüÁ¶). °Ô´Ù°¡ ´Ü¸éµµµéÀ» Â÷·Ê´ë·Î scanningÇÔÀ¸·Î½á ¼¼Æ÷¼Ò±â°üµéÀÇ 3Â÷¿ø ±¸Á¶¸¦ ¾Ë¾Æ³¾ ¼ö ÀÖ½À´Ï´Ù.
